Studies on the Impairment of Liver Function and the Changes of Coagulation Factors in Carbon Tetrachloride Intoxicated Rabbits


Abstract
It has been well known that hemorrhagic tendency appears among the patients with serious hepatic diseases, although much is still not known about the pathogenesis of this phenomenon.
In our laboratory, S. S. Lah observed the deficiencies of blood coagulation factors in the patients with various hepatic diseases, and compared these deficiencies with hepatic
functional disturbances and findings of liver biopsy in these patients. Then S. K. Kim, K. W. Kim, and T. W. Han investigated the deficiencies of blood coagulation factors, hepatic functional disturbances and histological changes in liver of rabbits intoxicated with phosphorus., alloxan and chloroform experimentally.
Therefore, the author treated rabbits with CCl4 (carbon tetrachloride), and at the same time, compared the findings on changes in the blood coagulation factors and the results of liver function tests and the histological changes of liver tissue.
The results were summarized as follows:
1) Changes in blood coagulation factors.
The time of blood coagulation prolonged markedly after the administration of CCl4. At the same time there occurred diminution of labile and stable factors, thromboplastin generating activities in serum factors and a marked prolongation of prothrombin time. 2) Liver function test.
There were decreased serum protein and A/G ratio, moderately increased serum cholesterol and degree of BSP retention and marked elevation of the serum transaminase activity after the administration of CCI4.
3) Histological changes in the liver.
The liver tissues showed central necrosis and fatty changes, and hydropic degeneration of liver parenchymal cells.
These coagulating, functional and histological changes in the groups treated with CCl4 6 times were more marked than in the groups treated 3 times.
